If you don't mind going a tad farther afield, I can recommend the Monfragüe campsite 8kms from Plasencia in the province of Cáceres, which is west of Madrid. Very friendly staff, decent showers, large pitches and generally quiet campsite with fairly good restaurant. More than adequate for one night, good base for spending more time in the area.
Monfragüe is about five hours from Gijón, which is west of Santander on the north coast, so it may take a tad longer to get to Monfragüe from Santander.
Just thought -seeing as you don't arrive in Santander till 6pm, you might want to try somewhere north of Madrid. If your route takes you through the province of León and anywhere near a little town called La Vecilla (about 20 mins from La Robla, I think) then try the La Cota campsite. Very nice people, small campsite, largish grassy pitches, well frequented by foreign campers (especially Dutch).
Me again..another afterthought.... you need to bear in mind that many campsites close their gates to vehicles at a certain time (usually 10pm but it varies from campsite to campsite)so perhaps aiming for west of Madrid from Santander as of 6pm and arriving in time is a bit ambitious.
I'm not sure what route you'll be taking from Santander but if you'll be travelling a little westwards from Santander along the north coast before you head south, then you have two interesting campsites in Cantabria. I've never been to either (might be going to one of them this weekend but I've heard they're both okay.. names:
-Camping El Rosal in San Vicente de la Barquera
-Camping Playa de Oyambre, which is half way between Comillas and San Vicente de la Barquera.

The Cantabrian ones will take you along the coast ... well off route for Madrid and south.
Salamanca has a reasonable 'town' site and the city is well worth a look ... the central square is rated the best in Spain ...
A bit further west is a good site 'Camping Sierra de Francia' not far from the historically interesting Cuidad Rodrigo.
Santander to Salamanca is about four hours by car ... longer of towing ...
You could book ahead and arrange for late arrival.
Campin El Eden, near Palencia looks fine and Camping Fuente de los Canos ... before Palencia looks good, too.
Kelper's right, going westwards along the north coast will take you off route if you're heading for Madrid, but not necessarily so if you're heading west of Madrid and ultimately to the Sevilla/Cádiz area. It depends what route you want to take and what you want to see/do on the way south. And of course, booking ahead for late arrival is an option, assuming the campsite will let you book ahead. We've been having problems finding campsites up here in Asturias that will actually take a booking this summer, but then we've got a tent and often campsites consider tenters as the poor relatives, so to speak, in peak season, .
Near the city of Salamanca, there are two campsites that I know of.. Camping Don Quijote, which I can't speak for, and Camping Ruta de la Plata, which is small and (back in 2008) was not the best of campsites but would be okay for one night or two. If I remember rightly, this one is somewhere near the Autovía Ruta de la Plata. And as Kelper says, Salamanca is well worth a visit.. not only the Plaza Mayor, but the two cathedrals (one of the best altars I've ever seen is in the old cathedral), the Casa de las Conchas (House of Shells, has shell shapes on the façade), the University building with its toad and skull.. etc etc..and then there's the lentils from Armuña..best lentils I've ever had, just lentils with nothing added, with a menu of the day...
My god, I think I should work for the Spanish Tourist Office!!

After arriving in Santander at 6pm ,how many hours do you want to drive and on which route.
Once you have decided on that evenings approx. destination, you can the use google maps to see where the Campings are located.
( eg: just type camping Burgos or camping Palencia. It will also give you the mileage from Santander if you fill in the directions box ).
